Big Mutha Truckers has players driving an 18-wheeler across various cities on their way to amassing a fortune hauling cargo. Rival truckers desperately want a piece of the action, of course, and players will also have to contend with law enforcement, biker gangs, and even disgruntled family members while earning a decent living. Rather than focus solely on the racing action as in games like 18-Wheeler: American Pro Trucker, Big Mutha Truckers emphasizes the business associated with selecting the best type of goods to haul and where to go to sell them at a profit. 

The game also has a storyline created with its tongue firmly in its cheek. Cast in the role of Ma Jackson's son (one of four, each fathered by a different man), players find themselves competing with their siblings to see who can pull in the most cash. Five cities are available to drive to, whose population may or may not be receptive depending on which brother is selected. Each city also has a store, a bar, and a garage. The store is where players select their cargo, while the bar is the place to receive high-interest loans from a loan shark, listen to gossip on the latest market trends, and apply for offbeat side jobs from the bartender.

Not surprisingly, the garage is the business of choice for refueling, repairing, and remodeling one's rig. Players will receive damage during their crossings into biker territory, whose leather-clad enforcers will try to hijack the truck in a manner reminiscent of The Road Warrior. To stop them, players can swerve their rig to sideswipe the motorcycles onto the asphalt. The garage also lets players modify their truck's logo or upgrade their carrying capacity (such as purchasing a tanker for hauling oil or other liquids). Developer Eutechnyx' previous experience in the racing genre includes 007 Racing, F1 World Grand Prix, and Test Drive Le Mans.
